What is Anti Asialo GM1 antiserum?

Anti Asialo GM1 antiserum is used in laboratory research and has been shown to eliminate natural killer (NK) activity in the cells of various strains of mice and rats. Anti Asialo GM1 antiserum reacts with:

 

- Mouse and rat NK cells

- Mouse monocytes (liver cells that contain no NK cells, bone marrow, fetal liver cells, spleen cells of nude mice macrophages)

- Fetal thymocytes (12 days old, with ratio of existence decreased gradually until there are none in newborn mice)

 

Anti Asialo GM1 lasts 2 years before reconstitution, and 2 to 3 months after reconstitution, when it is kept refrigerated. Anti Asialo GM1 lasts only two to three days at room temperature. Distilled water can be used for reconstitution. Since the material is lyophilized with salts, use of other solvents such as PBS or MEM may increase the salt concentration.

 

This antiserum should NEVER be frozen.

 

The following is information about injections using Anti Asialo GM1 antiserum:

 

Mouse-intravenously: 10 – 50 µL (approximately 20 µL). The exact dosage should be decided from the titration data enclosed with the package, and the exact nature of the study. The first injection may be effective for four days with gradual diminution. Therefore, three to four injections are necessary for a two week study.

 

Rat – intravenously: 50 – 250 µL (4 to 5 times the usual mouse dose is required.) Health conditions and the weight of rats should be taken into consideration. It is recommended that the researcher assay NK activity to determine the proper injection dosage.

 

Mouse and rat – intraperitoneally – Dosage should be equal to or greater than the IV dosage.
